干旱胁迫对转CpHRD基因烟草的影响

摘 要:

HRD基因是AP2/EREBP转录因子家族的一员, 过表达HRD基因可以提高水稻的水分利用效率。本文采用同源序列克隆的方法从线果芥中克隆了CpHRD基因, 并转化野生型烟草, 测定了干旱胁迫下转CpHRD基因烟草的丙二醛、游离脯氨酸、可溶性淀粉、可溶性糖的含量以及过氧化氢酶和超氧化物歧化酶活性6项生理指标; 观察了转基因烟草的形态和气孔的变化。结果显示, 除MDA含量外, 转CpHRD基因烟草的其余5项指标均显著高于野生型烟草, 而转基因烟草的叶表面气孔数和气孔开度均低于野生型, 说明CpHRD基因参与了植物逆境应答过程, 并可以显著提高转基因烟草的抗旱能力。

Influence of Drought Stress on Transgenic CpHRD Gene in Tobacco

Abstract:

HRD is a member of the AP2/EREBP transcription factor family, Overexpression of HRD in rice improved its water use efficiency (WUE). A CpHRD gene derived from Conringia planisiliqua was cloned by homology-based cloning method and transformed the tobacco (Nicotiana tabacum) leaf. The six physiological and biochemical indicators (the contents of MDA, PRO, H2O2, soluble starch, soluble sugar, and the activities of CAT, SOD) were measured in transgenic CpHRD tobacco under drought stress. And the changes of morphology and porosity of transgenic tobacco were observed. The results indicated that expect the MDA contents, the other five indicators in transgenic tobaccos were significantly higher than those in the wild type tobacco. The stomata number and stomata aperture of transgenic tobacco leaves were lower than those of the wild type. These results also indicated CpHRD gene involved in abiotic stress responses and strengthen the drought resistance of transgenic tobaccos.
